MBX Biosciences Announces U.S. FDA Clearance of IND Application for MBX 1416 for the Treatment of Post-Bariatric Hypoglycemia

Phase 1 trial initiation anticipated Q3 2023; Topline results expected in 2H 2024

CARMEL, Ind., August 7, 2023MBX Biosciences, Inc., a clinical stage biopharmaceutical company developing Precision Endocrine Peptide™ (PEP™) therapeutics to treat an array of endocrine disorders, today announced that the U.S. Food and Drug administration (FDA) has cleared the Investigational New Drug (IND) application for MBX 1416, the company’s long-acting glucagon-like peptide 1 (GLP-1) receptor antagonist, being developed for the treatment of post-bariatric hypoglycemia (PBH).

MBX 1416 is an investigational long-acting GLP-1 receptor antagonist in development as a potential treatment for PBH. It was designed using the company’s novel, proprietary PEP platform to prevent the occurrence of severe hypoglycemia in individuals with PBH so they can lead healthier and more independent lives. MBX expects to initiate enrollment in a Phase 1 trial in the third quarter of 2023 and anticipates reporting topline results in the second half of 2024.

About Post-Bariatric Hypoglycemia

Post-bariatric hypoglycemia (PBH) is a rare disease and a serious complication of bariatric surgery. PBH is characterized by repeated episodes of symptomatic hypoglycemia, triggered by excessive insulin levels following a meal, and generally presents as early as six months after Roux-en-Y gastric bypass (RYGB). Hypoglycemic episodes can occur multiple times per day and can periodically manifest with severe symptoms such as dizziness, confusion, loss of consciousness or seizure due to glucose deficiency in the brain. The unpredictability of hypoglycemic episodes and their associated risks may meaningfully hinder daily activities. As a result, the patient burden can be substantial, and many individuals cannot drive, work or live alone. To date, there are no approved pharmacotherapies to treat PBH. As the use of surgery to address metabolic conditions continues to rise, the incidence of PBH is expected to increase, reinforcing the need for safe and effective therapies.

Dr. Prestrelski is a seasoned biopharmaceutical executive with over 30 years of scientific and operational expertise who has led successful endocrine and metabolism product development programs from discovery through regulatory approval.

The Series B financing was led by Wellington Management with participation from RA Capital Management and Norwest Venture Partners along with existing investors Frazier Life Sciences, New Enterprise Associates (NEA) and OrbiMed. In addition, Tiba Aynechi, Ph. D., general partner at Norwest Venture Partners, will join the MBX board.

About Hypoparathyroidism

Hypoparathyroidism (HP) is a rare endocrine disorder caused by a deficiency of parathyroid hormone that results in decreased calcium and increased phosphorus levels in the blood. HP affects approximately 200,000 individuals worldwide, most of whom develop the condition following damage to or removal of the parathyroid glands during thyroid surgery. It is a complicated disorder associated with a wide range of symptoms including paresthesias, muscle cramps, seizures, an impaired quality of life, and an increased risk of comorbidities including kidney stones and impaired renal function. The goal of treatment is to maintain blood calcium levels in the low-normal range while preventing symptoms of hypocalcemia. MBX 2109 is in clinical development for treatment of hypoparathyroidism

CARMEL, Ind., Oct. 05, 2022 — MBX Biosciences, Inc., a clinical stage biopharmaceutical company developing Precision Endocrine Peptide™ (PEP™) therapeutics to treat an array of endocrine disorders, today announced that it has initiated the multiple ascending dose portion of its ongoing Phase 1 clinical trial of MBX 2109. MBX 2109, the company’s lead investigational drug, is a long-acting parathyroid hormone (PTH) peptide prodrug in development for the treatment of hypoparathyroidism.

“This is an exciting achievement for MBX as we continue to advance our first clinical program,” said Kent Hawryluk, president and chief executive officer of MBX Biosciences. “We believe MBX 2109 has the potential to address a substantial need in hypoparathyroidism, and this trial brings us a step closer to evaluating it in patients.”

The randomized, double blind, placebo-controlled Phase 1 trial is designed to evaluate the safety, tolerability, pharmacokinetics and pharmacodynamics of single and multiple ascending doses of MBX 2109 in healthy adult subjects. The single ascending dose portion of the trial, which began at the end of 2021, was completed successfully. MBX 2109 was observed to be safe and well tolerated, as compared to placebo. For the multiple ascending dose phase of the trial, subjects are to receive either MBX 2109 or placebo once weekly subcutaneously for four weeks. The primary outcome measures will evaluate safety and tolerability. Secondary outcome measures will evaluate pharmacokinetics and pharmacodynamics. CARMEL, Ind., July 26, 2022 — MBX Biosciences, Inc., a clinical stage biopharmaceutical company developing precision endocrine peptide therapeutics to treat endocrine disorders, today announced that the U.S. Food and Drug Administration (FDA) granted Orphan Drug designation to its lead therapeutic product candidate, MBX 2109, for the treatment of hypoparathyroidism.

Orphan drug designation is designed to support the development of medicines addressing rare medical diseases or conditions that affect fewer than 200,000 people in the United States. MBX 2019 is an investigational long-acting parathyroid hormone (PTH) prodrug. MBX Biosciences Appoints Richard B. Bartram as Chief Financial Officer

CARMEL, Ind., April 11, 2022 – MBX Biosciences, Inc., a clinical stage biopharmaceutical company developing peptide therapeutics with a focus on the treatment of endocrine disorders, today announced the appointment of Richard B. Bartram, as Chief Financial Officer. Mr. Bartram is a proven financial executive with an extensive understanding of the capital markets and a wide variety of experience in private transactions, initial public offerings, follow-on equity offerings, debt offerings, and other financial transactions.

Most recently, Mr. Bartram served as Chief Financial Officer at Esperion Therapeutics Inc., a publicly traded pharmaceutical company focused on the development and commercialization of therapies for patients with elevated LDL cholesterol. During his time at Esperion, he led the company’s successful efforts to raise more than $1.2 billion in capital, supporting the approval and commercialization of both NEXLETOL® and NEXLIZET®. In addition, he provided key contributions to several successful business development licensing transactions. Prior to his tenure at Esperion, Mr. Bartram served as an assurance manager with PricewaterhouseCoopers LLP from 2005 to 2013. He earned his undergraduate and graduate degrees in Accounting from Michigan State University, and is a Certified Public Accountant.
